Western blot was performed using Anti-Follistatin Polyclonal Antibody (Product # PA5-79284) and a 34 kDa band corresponding to Follistatin was observed across the panel tested upon treatment with Protein Transport Inhibitor Cocktail (PTI) (Fig.a) and upon glucose deprivation in HeLa cells (Fig.b). Whole cell extracts (30 µg lysate) of A549 (Lane 1), A549 treated with PTI (1X for 4 hr) (Lane 2), SK-O-V3 (Lane 3), SK-O-V3 treated with PTI (1X for 4 hr) (Lane 4) (Fig.a) and nuclear enriched extract ( 30 µg lysate) of HeLa (Lane 1) and HeLa (glucose deprived for 12 hr) (Lane 2) (Fig.b) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.2 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
